As a beta-lactam antibiotic, Longocilline works by interfering with the synthesis of the bacterial cell wall. It binds to specific proteins within the cell wall, preventing the bacteria from maintaining their structural integrity. This process leads to the eventual destruction of the targeted bacteria. Because of its formulation, a single administration can maintain therapeutic levels of the antibiotic in the body for several weeks, depending on the specific condition being addressed.
Therapeutic Applications
Longocilline is primarily used to treat infections caused by bacteria that are highly sensitive to penicillin. Its long-acting nature makes it particularly useful in specific clinical scenarios where sustained antibiotic presence is required. Common applications include:
- Syphilis: It is used in the treatment of various stages of syphilis, an infection caused by the bacterium Treponema pallidum.
- Rheumatic Fever Prevention: It is utilized as a preventive measure (prophylaxis) against recurrent attacks of rheumatic fever by eliminating streptococcal bacteria that can trigger the condition.
- Respiratory Infections: In certain cases, it may be used for the treatment of specific streptococcal infections of the upper respiratory tract, such as tonsillitis or pharyngitis.
Spectrum of Activity
Longocilline is effective against a variety of Gram-positive organisms and some Gram-negative cocci. It is specifically active against:
- Streptococcus pneumoniae
- Groups A, C, G, H, L, and M streptococci
- Treponema pallidum (the causative agent of syphilis)
- Corynebacterium diphtheriae
