Clofibrate is a lipid-regulating agent that belongs to the fibrate class of medications. It was developed to assist in the management of high serum lipid levels, specifically targeting cholesterol and triglycerides circulating in the bloodstream.
Mechanism of Action
The medication works by increasing the activity of lipoprotein lipase, an enzyme responsible for breaking down triglycerides. By enhancing this process, clofibrate helps reduce the concentration of very-low-density lipoproteins (VLDL) and low-density lipoproteins (LDL), while potentially causing a modest increase in high-density lipoprotein (HDL) levels, often referred to as "good" cholesterol.
Historical Context and Usage
Clofibrate was one of the first fibrates used extensively in clinical practice. It was primarily indicated for individuals with specific types of hyperlipidemia who did not respond adequately to dietary changes alone. While it served as a foundational treatment in cardiovascular pharmacology, its use has largely been superseded by newer fibrates and other classes of lipid-lowering medications that offer different metabolic profiles.
Clinical Application
In a clinical setting, clofibrate was used to treat:
